I know the showerhead unscrews from the wall fitting but will I need to turn off the water supply before I do this?

Just make sure the shower is turned off

No the flow of water is controlled by the shower valve not the head so you you will be OK. It's always wise however to isolate the water supply to the shower before performing any maintainance tasks.
